SUBSTANCE: reagent used for determination of hemoglobin content and amount of leukocytes in blood sample involves at least one detergent of cationic type; saponin; at least one inorganic salt and osmotic and/or leukoprotective agent; and organic and/or inorganic buffer suitable for selective adjustment of pH value of reagent being at practically neutral pH value 5-8 or basic pH value 8-12. Using reagent allows carrying out the determination of hemoglobin content without cyanide using and to identify and determine amount of leukocyte subpopulations.
