THERMOSTABLE SULFOLOBUS SOLFATARICUS GLUCOAMYLASE AND METHOD FOR PREPARING GLUCOSE USING THE SAME

PURPOSE: Thermostable Sulfolobus solfataricus glucoamylase and a method for preparing glucose using the same are provided, thereby producing glucose without a cooling process because the Sulfolobus solfataricus glucoamylase can be used at high temperature. CONSTITUTION: Thermostable glucoamylase is produced from Sulfolobus solfataricus, has optimal pH of 5.0 to 6.5 and optimal temperature of 70 to 95 deg. C, and decomposes α-1,4 glucoside bond and α-1,6 glucoside bond, wherein the substrate of the thermostable Sulfolobus solfataricus glucoamylase includes maltose, maltotriose, maltotetraose, maltopentaose, maltohexaose, maltoheptaose, isomaltose and starch. A method for preparing glucose comprises the steps of: treating starch with α-amylase to obtain a liquified reaction solution; and treating the liquified reaction solution with thermostable Sulfolobus solfataricus glucoamylase and thermostable pullulanase.
