PURPOSE: A Tat-PTEN fusion protein and a process for preparing the same and the use thereof are provided. The Tat-PTEN fusion protein penetrates subconjunctival fibroblast and inhibits TGF-β(transforming growth factor-β) signal transmission. Therefore, the apoptosis of the subconjunctival fibroblast can be induced and the formation of a scar can be inhibited. CONSTITUTION: A Tat-PTEN fusion protein contains PTEN(phosphatase and tensin homologue deleted from chromosome 10) protein and protein transduction domain (SEQ ID NO:1) of HIV-1 Tat protein bound to the amino terminal of the PTEN protein, wherein the Tat-PTEN fusion protein has the amino acid sequence set forth in SEQ ID NO: 12. A recombinant polynucleotide comprises a DNA sequence encoding the Tat-PTEN fusion protein, wherein the recombinant polynucleotide has the nucleotide sequence set forth in SEQ ID NO: 11. A vector expressing the Tat-PTEN fusion protein contains the recombinant polynucleotide, wherein the vector has a restriction enzyme map shown in figure 3. A process for preparing the Tat-PTEN fusion protein comprises expressing the vector in a microorganism and purifying the expressed Tat-PTEN fusion protein, wherein the microorganism is E. coli. |
