METHOD FOR CONSTRUCTING CODING REGION ENRICHED GENOMIC LIBRARY

PURPOSE: A method for constructing coding region enriched genomic library is provided, thereby carrying out partial denaturation and cutting by S1 nuclease more effectively in the noncoding region than the coding region. CONSTITUTION: A method for constructing coding region enriched genomic library comprises the steps of: (i) treating genomic DNA with S1 nuclease to partially denaturate the genomic DNA; (ii) treating the partial denatured genomic DNA with S1 nuclease; (iii) blunting both ends of the genomic DNA fragments; and (iv) inserting the genomic DNA fragments with blunt ends into a plasmid, wherein the blunting of both ends is carried out by T4 DNA polymerase.
