PROBLEM TO BE SOLVED: To obtain a leak-free membrane by optimizing a method for including an enzyme, especially a cytochrome c oxidase, in phospholipid vesicles. SOLUTION: The membrane is obtained by the following process. The cytochrome oxidase and phospholipids are dispersed in an aqueous solution and the resulting dispersion is dialyzed with a buffer solution as the outer liquid under specific conditions, wherein the concentration of the buffer solution is gradually lowered at least in three steps within the range of 0.1 to 300 mM and after the concentration comes to a minimum, the concentration is brought to a higher level. When the mixed liquid of the enzyme solubilized by a surfactant and the phospholipids is dialyzed under the above conditions, the concentration of the surfactant is gradually lowered, the solubility of the enzyme lowered, resulting in including the enzyme in the phospholipid vesicles. The hydrogen ion transport activity of the phospholipid vesicles obtained above remains constant after addition of a reduced cytochrome c as shown in the figure 3, meaning that the membrane is hard to permeate hydrogen ions.
