PURPOSE:To efficiently obtain the subject compound, useful as a synthetic raw material for medicines, food additives, etc., by reacting an enzymic source capable of catalyzing the hydroxylating reaction at the 3-position of L-proline with bivalent iron ions, 2- ketoglutaric acid and the L-proline in an aqueous medium. CONSTITUTION:This method for producing cis-3-hydroxy-L-proline is to react an enzymic source which is a culture, a microbial cell, a treated substance of the microbial cell, a purified enzyme or a crude enzyme of a microorganism, belonging to the genus Streptomyces or Bacillus, and manifesting the following properties: having functions to hydroxylate the 3-position of L-proline in the presence of 2-ketoglutaric acid and bivalent iron ions and 6.5-7.5 optimum pH by reaction at 30 deg.C for 20min; stable at pH6.5-8.0 by treatment at 4 deg.C for 23hr; having 35-40 deg.C optimum temperature by reaction at pH7.0 for 15min; inactivated by treatment at pH7.5 and 50 deg.C for 30min and having 35000+ or -5000 molecular weight [measured by the sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS)-PAGE)] and 4.3 isoelectric point as an enzymic source for catalyzing the hydroxylating reaction of the 3-position of the L-proline with the bivalent iron ions, 2-ketoglutaric and L-proline in an aqueous medium. Thereby, the objective compound is industrially and advantageously obtained. |
