MICROORGANISM BACILLUS SP. WL-7 PRODUCING MANNANASE

PURPOSE: A microorganism Bacillus sp. WL-7 producing mannanase is provided. The microorganism being similar to Bacillus subtilis has improved stability and mass-produce mannanase, and the produced mannanase has higher reactivity at neutral pH and medium temperature, and improved hemicellulose-degrading activity. CONSTITUTION: A microorganism Bacillus sp. WL-7(KCTC 10279BP) isolated from soybean paste, having optimal activity at pH 5.0 to 7.5 and 37 deg. C and producing mannanase which degrades hemicellulose in crops is provided. A method for mass-producing mannanase comprises the steps of: adding 0.5% of Konjac(or Devil's tongue) and 0.5% of locust bean gum(LBG) into an LB medium containing 10 g/l of tryptone, 5 g/l of an yeast extract and 5 g/l of NaCl to prepare a medium for Bacillus sp. WL-7(KCTC 10279BP); inoculating Bacillus sp. WL-7(KCTC 10279BP) into the medium and culturing it at 37 deg. C for 6 to 15 hours; concentrating the supernatant of the cultured medium; fractioning the supernatant with ammonium sulfate salt to obtain a precipitate; diluting the precipitate in 50 mM of a sodium phosphate buffer solution(pH 6.5); and dialyzing the dilution with 50 mM sodium phosphate buffer solution(pH 6.5).
