The present invention relates to a method for constructing a linear expression vector. Particularly, the present invention provides a method for constructing a linear expression vector which includes: amplifying gene fragments by using (DNA+RNA) hybrid as primer, and ligating each fragment by subsequent step of digesting RNA. The present invention is not affected by sequence change to target DNA fragment. The linear expression vector constructed as above can be used to transform host cell to express target protein, and provide technical platform for fields of gene expression, production of antibody and vaccine, and research of protein function.
