The present invention is to provide polypeptide fine particles which comprise adding a polypeptide-insoluble water-miscible organic solvent to a frozen product of an aqueous solution containing a polypeptide and a phase separation inducer, and dissolving the separation inducer and ice in the frozen product; a polypeptide fine particle dispersion obtainable by the method; polypeptide fine particles obtainable by removing the phase separation inducer, water and water-miscible organic solvent from the polypeptide fine particle dispersion; a method of producing microspheres which comprises dispersing the polypeptide fine particles in a solution in which a biodegradable polymer is dissolved in a volatile and water-miscible organic solvent, emulsifying the mixture in an aqueous phase to prepare an O/W emulsion, and removing the water-miscible organic solvent; and a microsphere preparation obtainable by the method. <IMAGE>
