Preparing a maize callus culture (A), comprises: (a) recovering leaf tissue from the coleoptile region of a seedling; and (b) inducing callus formation by culturing the tissue in the presence of proline, silver nitrate and at least one auxin, is new. Independent claims are also included for: (1) producing a maize suspension culture by conventional conversion of (A); (2) preparing transgenic maize callus, comprising suspension or protoplast cultures by standard processes, but including subjecting (A) to transformation; (3) producing transgenic maize plants, or their parts, by subjecting leaf tissue (from step (a)) or the new callus to transformation then regeneration; (4) preparing optionally transgenic maize plants, or their parts, by regeneration of (A); and (5) transgenic maize plants, or their parts, produced by the method of (4).
