PROBLEM TO BE SOLVED: To provide a method and a device for separating and collecting a cell suitable for cell diagnosis easily, in a short period of time, and efficiently, and provide a nucleated cell containing composition obtained by the method and the device. SOLUTION: The method of preparing a sample for cell diagnosis is characterized in that it introduces a nucleated cell containing liquid into a nucleated cell trapping means, which traps a nucleated cell, and does not substantially trap a nonnucleated cell, and collects the nucleated cell trapped in the nucleated cell trapping means with maintaining the shape of the cell, after allowing the nucleated cell to be trapped, and allowing the nonnucleated cell to be flushed away. The cell separating device therefor comprises a nucleated cell trapping means (1), which traps a nucleated cell, and does not substantially trap a nonnucleated cell, a nucleated cell containing liquid injecting means (2), a liquid discharging means (3), a collected liquid injecting means (4), a nucleated cell collecting means (5), and a cleaning liquid injecting means (6).
