Enzymes having an activity of transferring N-acetylgalactosamine via an &agr;1-bond to a hydroxyl group of serine or threonine in a core protein or a peptide sequence are isolated and the structures of genes thereof are clarified, thereby enabling the genetic engineering production of these enzymes and so on, the production of GalNAc-modified peptides, GalNAc-modified proteins, etc. as substrates based on these enzymes, the identification of a protein having an unknown O-bonded sugar chain attached thereto or the O-bonded sugar chain modification site of an unknown peptide based on these enzymes, and diagnosis of diseases based on these genes. Use is made of proteins respectively having the amino acid sequences represented by SEQ ID NOS: 1 to 7, amino acid sequences derived therefrom by substitution or deletion of one or more amino acids or amino acid sequences derived therefrom by insertion or addition of one or more amino acids and having an activity of transferring N-acetylgalactosamine (GalNAc) via an &agr;1-bond to a hydroxyl group of serine or threonine in a core protein or a peptide serving as a substrate, and nucleic acids encoding these proteins.
