PURPOSE: A method for detecting electrochemiluminescence of nucleic acid using an intercalator is provided to efficiently detect complementary double-stranded nucleic acid by using the intercalator combined with a hybridized nucleic acid. CONSTITUTION: A thiol functional group is fixed on a surface of a working electrode by using a nucleic acid solution. The surface of the working electrode is cleaned by a phosphoric acid solution and is sunk in a buffer solution including a target nucleic acid having a basic sequence complementing the nucleic acid solution to execute hybridization. A solution obtained by melting an intercalator in a buffer is added to the hybridized nucleic acid to combine the intercalator with the hybridized double-stranded nucleic acid. An uncombined intercalator is removed by cleaning the working electrode with the buffer solution. The working electrode is put on a measuring container and a transition metal complex is added before the working electrode, a reference electrode, and a counter electrode are installed. Voltage is applied to the working electrode so that the intercalator combined with the double-stranded nucleic acid induces an oxidation-reduction reaction to appear electrochemiluminescence. |
