<p>Immunoassay or nucleic acid hybridisation assay for qualitative or quantitative determn. of an analyte in a sample uses the following components: (a) a capture reagent having two binding sites, one of which permits (opt. together with other assay components) specific detection of the analyte and the other of which can be bound to an active solid phase through a specific binding pair (sbp), where one member of the sbp is bound to the capture reagent and the other member is coupled to the active solid phase; (b) an active solid phase, and (c) an inactive solid phase which is the same as the active solid phase except that the capture reagent cannot bind to it. The sample is first contacted with the inactive solid phase and only later contacted with the active solid phase, or the sample is contacted with the active and inactive solid phases simultaneously. The capture reagent and opt. other assay components are either present from the start or are added later, where upon determn. of the analyte takes place. Also claimed is a method for avoiding interference from non-specific binding to the solid phase in an assay as above, comprising incubating the sample in the presence of a first active solid phase, removing the sample from the first phase, and incubating the sample in the presence of the capture reagent (and opt. other assay components) and a second, largely identical, active solid phase.</p> |
