PROBLEM TO BE SOLVED: To clone a testosterone metabolic pathway system enzyme gene in C. testosteroni TA441 strain, to produce the gene-broken strain, and to analyze its structure. SOLUTION: The testosterone metabolic pathway system enzyme gene is cloned, and its structure is analyzed. Further, the cloned enzyme gene is used to produce the gene-broken strain in which the enzyme gene is broken. A method for using the gene-broken strain to produce a testosterone metabolic intermediate is provided. The gene-broken strain is a gene-broken strain which participates in the testosterone metabolic enzyme and in which a gene encoding either of five specific amino acids existing on the chromosomal DNA of a microorganism cell or encoding either of the amino acids wherein one to several amino acids are deleted, substituted, added and/or inserted and which has the testosterone metabolism activity is broken.
