Use of the regulatory subunit of the camp dependent protein kinase (PKA) from dictyostelium for camp measurements.

The use of a DNA construct for inducing the expression of a fusion protein in a host cell for the measurement of camp concentration is described. The DAN construct comprises the coding sequence of at least one camp binding site of the regulatory subunit (R) of a cAMP dependant protein kinase that is unable to dimerise which coding sequence is operably linked to a DNA sequence encoding a reporter polypeptide. Preferably the camp dependent protein kinase is from Dictyostelium discoideum. Also described are methods for measuring the cAMP concentration in a biological fluid using a fusion protein encoded by the construct and methods for measuring cAMP within living cells excluding human cells in situ using the constructs
