PURPOSE: Provided are a bio-device in which a testing substance in a sample solution can be quantitative-measured promptly and with high accuracy and superior reproducibility, a quantitative measuring device using the same and a quantitative measuring method. CONSTITUTION: In a bio-device for measuring a testing substance included in a test solution, the bio-device comprises a sample introduction part, a detected substance holding part and a determination part. The sample introduction part, the detected substance holding part and the determination part are arranged so that the sample solution introduced into the sample introduction part moves to the determination part through the detected substance holding part, and at least the detected substance holding part and the determination part are constituted by the same member. A first substance group containing a substance which reacts particularly on the test substance is included in the detected substance holding part in a state of being able to elute with the sample solution, and after the first substance group has been eluted from the sample introduced into the sample introduction part, the first substance group is spread as an agglomeration having a start end and a terminal end in elution and spread processes.
