PROBLEM TO BE SOLVED: To provide a method and a composition relating a ribonucleoprotein enzyme involved in human telomere DNA synthesis to the fields of molecular biology, chemistry, pharmacology and medical and diagnostic technology. SOLUTION: This isolated or synthetic, substantially pure or recombined polynucleotide comprises at least 10 consecutive nucleotides which are identical or complementary to a contiguous sequence of a human telomerase RNA component gene contained in an about 2.5 kb HindIII-SacI insert of plasmid pGRN33 (ATCC 75926) and the polynucleotide is not a dimer or other polymers of 5'- TTAGGG-3'. Furthermore, the polynucleotide is (a) a probe secifically hybridizing to the plasmid ATCC 75926 insert, (b) one of a set of primers specifically amplifying a part of the plasmid ATCC 75926 insert or (c) a polynucleotide inhibiting the activity of telomerase reverse transcriptase.
