The invention relates to methods and agents for controlling bone formation by modulating the ANK-pyrophosphate channel and/or simulating the function thereof. The invention can be used in the field of human medicine, particularly for illnesses whose cause stems from a disturbed balance between bone formation and degradation such as osteoporosis. The invention defines the human ANK-protein as the main protein in the development of medicaments ("drugable target") for therapy and prophylaxis of osteoporosis. The invention also relates to novel human ANK-gene-variants and the use thereof.
