PURPOSE: Provided are a kit for detecting the nucleic acid of Mycobacterium tuberculosis with specificity and sensitivity and a detection method of Mycobacterium tuberculosis by using the kit. CONSTITUTION: The detection kit comprises a DNA extract solution containing weak acid ion exchange resin and a PCR master mixed solution including distilled water, buffer solution, dNTP, 2 kinds of primer sets and a mixed enzyme solution of TaqDNA polymerase and uracil DNA glycosylase, wherein the 2 kinds of primer sets have nucleotide sequences of SEQ ID NO:1 and 2, and SEQ ID NO:3 and 4. The detection method comprises the steps of: treating a sample with a DNA extract reagent and a PCR master mixed solution then performing PCR; and analyzing the presence of Mycobacterium tuberculosis on the gel.
