PURPOSE: A screening method for the detection of multidrug resistance protein inhibitors and reactive oxygen species-producing cytotoxic substances using the doxorubicin-resistant acute myelocytic leukemia subline AML-2/DX10 is provided, therefore novel multidrug resistance protein inhibitors can be easily found in nature. CONSTITUTION: The method for screening multidrug resistance protein inhibitors comprises the steps of: culturing AML-2/DX100 in a medium without drugs for 4 to 6 days; adding doxorubicin into the AML-2/DX100 cultured medium; inserting each material to be screened into each well of a 96-well microplate; adding the doxorubicin treated AML-2/DX100 cultured medium into each well; incubating the 96-well microplate in a CO2 incubator for 3 days; adding an MTT solution into each well and incubating it at 37 deg. C and 5% CO2 for 5 to 6 hours; adding HCl-isopropanol solution into each well and determining optical density of each well using a microplate reader.
