SUBSTANCE: invention describes a method of modification of (poly)peptides involving the incorporation of at least one specifically cleaved methionine and a label (tega) this is a His-label by the end of (poly)peptide chain in the process of its synthesis and protection of methionine residues in this (poly)peptide from cleavage by means of protection with sulfoxide group. Invention describes also a method of synthesis of the purified (poly)peptides involving the following stages: a) synthesis of the end peptide; b) attachment of at least one specifically cleaved methionine by the end of this (poly)peptide while methionine (methionines) in (poly)peptide are protected against cleavage by means of protection with sulfoxide group; c) elongation of this (poly)peptide and methionine (methionines) attached to it with a label to yield the elongated polypeptide; d) purification of the elongated (poly)peptide by method that is specific to a label; e) removal of label and additional methionine (methionines) from the elongated (poly)polypeptide by means of cleavage with cyanocyanocyanogen bromide to yield the purified (poly)peptide being the label means the following His-sequence: His-His-His- His- His-His-Gly-Gly-.
