PURPOSE: Bacillus sp. HSB-21 strain isolated from soil and chitosanase produced therefrom are provided. The produced chitosanase has a high production yield, is easily concentrated and used for the manufacturing of highly polymerized oligosaccharide. CONSTITUTION: The chitosanase is produced by the steps of: seed-cultivating Bacillus sp. HSB-21(KFCC-11221) in LB broth medium at 25 to 35 deg.C; inoculating the Bacillus sp. HSB-21 cultivated medium into LB medium and seed-cultivating it at 25 to 45 deg.C with agitation of 100 to 300 rpm; and inoculating the cultivated medium into a medium containing 0.5 to 3% of carbon source and 0.5 to 4% of nitrogen source under conditions of 25 to 45 deg.C, 6.5 to 7.5 pH and 1vvm with agitation of 100 to 300 rpm, in which the carbon source is selected from glucose, fructose, mannose, sucrose, trehalose, ribose, dextrin, soluble starch, chitosan and chitin; and the nitrogen source is selected from NH4Cl, NH4NO3, (NH4)2SO4, KNO3, yeast extract, peptone, tryptone, corn precipitate, beef extract and polypeptone.
