<p>A method of constructing a DNA library which comprises: (a) ligating unit domain DNAs in arbitrary combinations; (b) mixing the ligated unit domain DNAs thus obtained in such a manner that the terminal sequence of each of the ligated unit domain DNAs employed overlaps the terminal sequence of at least another type of ligated unit domain DNA; and (c) performing polymerase chain reaction by using the ligated unit domain DNA mixture as a template to thereby obtain a DNA library involving 2 or more clones. According to this method, a method of constructing a mass of genes wherein a plural number of DNA fragments encoding arbitrary amino acid sequences are ligated together in various orders at various lengths is provided.</p> |
