摘要 PROBLEM TO BE SOLVED: To provide a method for the determination of a DNA base sequence enabling the determination of the base sequence of a long DNA which has hitherto been difficult to determine. SOLUTION: The base sequence of a double-stranded DNA 4 is determined by (1) a step to bond an oligomer 6 having the recognition sequence of a class IIs restriction enzyme to a terminal of a double-stranded DNA 4, (2) a step to cleave the double-stranded DNA 8 having the oligomer bonded by the step (1) with a class IIs restriction enzyme, (3) a step to bond the oligomer to the fragment of the double-stranded DNA cleaved by the step (2) and (4) a step 20 to repeat the step (2) and the step (3) multiple times to obtain a series of double-stranded DNA fragments 9 composed of several number of bases and having a definite base length difference between the fragments. The present process enables easy determination of the base sequence of a long DNA fragment.
