PROBLEM TO BE SOLVED: To provide an analysis method for nucleic acid sequence, capable of readily detecting the kind of a base polymorphism in a specific nucleic acid sequence containing a mono base polymorphism contained in a specimen. SOLUTION: This method detects a mono base polymorphism on a sequence of a chromosome or its fragment. The method for detecting the kind of a base to be specified comprises reacting a reaction solution comprising a nucleic acid primer containing a base adjacent to a polymorphism base at the 3' end, any one of dideoxynucleotide corresponding to a polymorphism base having an identifiable characteristic or its mixture, a DNA polymerase and a composition necessary for activity expression of the DNA polymerase with a specimen nucleic acid and detecting the presence of incorporation of the dideoxynucleotide into the nucleic acid primer.
