PROBLEM TO BE SOLVED: To provide a cholesterol measuring sensor with only a slight variation in quality, capable of being easily and inexpensively manufactured without decreasing the measurement accuracy by solving a conventional problem. SOLUTION: The cholesterol measuring sensor forms an electrode system with at least a measurement electrode and a counter electrode on an insulative substrate and electrochemically detects a change in the material concentration of an enzyme, an electron acceptor, and a sample solution at reaction time by the electrode system so as to measure the substrate concentration of the sample solution. The above sensor is characterized as follows: the first layer is formed by retaining lipoprotein lipase, cholesterol esterase, cholesterol oxidase, and a surfactant on the measurement electrode and placing a retention body, through which the sample solution can pass, thereon; and the second layer is formed by retaining the surfactant on the counter electrode and placing the retention body, through which the sample solution can pass, thereon.
