<p>A vector for the cloning and expression of a polypeptide-encoding polynucleotide, which vector comprises: (a) a promoter; (b) a ligation-independent cloning (LIC) site; (c) at least one nucleic acid sequence encoding a tag; and (d) a nucleic acid sequence encoding a first reporter polypeptide, wherein a polypeptide-encoding polynucleotide cloned into the LIC site, when expressed, is expressed as a fusion polypeptide with the tag(s) and transcription of a polypeptide-encoding polynucleotide cloned into the LIC site is coupled to transcription of the nucleic acid sequence encoding a first reporter polypeptide.</p> |
