PURIFICATION OF HUMAN CILIARY NEUROTROPHIC FACTOR(CNTF) EXPRESSED FROM RECOMBINANT YEAST

PURPOSE: A purification of human ciliary neurotrophic factor(CNTF) expressed from a recombinant yeast is provided, thereby preparing a large quantity of high purity human CNTF. CONSTITUTION: A purification of human ciliary neurotrophic factor(CNTF) expressed from a recombinant yeast comprises the steps of: (a) crushing a recombinant yeast and centrifuging it to obtain an insoluble protein; (b) dissolving the insoluble protein in a buffer solution containing triton X-100 and centrifuging the solution; (c) diluting the supernatant thereof with a tris buffer solution; (d) subjecting the diluted solution to anion exchange resin column chromatography to obtain an eluate containing the protein; (e) dialyzing the eluate in the tris buffer solution; and (f) centrifuging the dialyzed human ciliary neurotrophic factor(CNTF) solution and subjecting the supernatant thereof to hydrophobic column chromatography.
