A test assembly for determination of blood cholinesterase activity comprises a first container 10 having therein a predetermined quantity of a colour change indicator responsive to pH changes and a second sealed container 241 containing a predetermined quantity of a stabilized anhydrous solution containing acetylcholine, the containers being interengageable so that the contents of one container may be mixed with the contents of the other. The blood sample is collected in capillary (26) of a first capillary pipette and added to the indicator solution in flexible-walled container 10 by compressing the container, inserting the pipette so that its end coupling (30) engages the neck of the container 10, and releasing the pressure on the container. The acetylcholine solution from container 241 is added to the contents of container 10 by breaking off the tip 27 of capillary 261, removing sealing plug 42, and engaging the capillary coupling with the neck of the container as described above. In the Examples, container 10 contains bromthymol blue (31 311-dibromothymol sulphonephthalein) and the acetylcholine solution is acetylcholine chloride in ethylene glycol. Other indicators are phenol red (phenol sulphonephthalein) and m nitrophenol. Acetylcholine iodide, bromide or perchlorate may be used instead of the chloride, and the solvent may be propylene glycol or dioxane.
