PURIFICATION OF STREPTOMYCIN

Streptomycin in a mineral acid salt form is purified from a fermentation broth containing streptomycin by (a) adsorbing the whole fermentation broth on a bed of a carboxylic acid cation exchange resin in an alkali metal salt form; (b) washing the streptomycin-containing resin bed with an aqueous solution of a chelating agent; (c) washing this bed with an aqueous solution of carbonic acid until substantially all alkali metal ions are removed therefrom, the streptomycin remaining on the resin bed through these steps .(a), (b) and (c); (d) eluting the purified streptomycin from the resin bed in a mineral acid salt form with a mineral acid solution of 1 to 5 N at a controlled flow rate, the mineral acid portions of the streptomycin-acid salt corresponding to the mineral acid used; and (e) recovering the streptomycin-mineral acid salt from the eluate. The chelating agent may be ethylenediamine tetra-acetic acid or nitrilotriacetic acid and the mineral acid may be sulphuric, nitric or phosphoric acid. The carboxylic acid cation exchange resin may be of the acrylate or methacrylate/styrene/divinyl benzene type preferably in the sodium salt form.
