RAT'S SKIP (SAPK INTERACTING PROTEIN) SELECTIVELY INHIBITING STRESS-ACTIVATED PROTEIN KINASE AND GENE THEREOF

PURPOSE: Provided is a SKIP(SAPK interacting protein), which selectively inhibits stress-activated kinase(SAPK). And its coding gene is also provided. Thereby, cell death is inhibited or sustained. The SKIP is used in treating cell death related diseases such as cancer, inflammation, apoplexy and the like, by controlling its functions. CONSTITUTION: SKIP(SAPK interacting protein), which selectively inhibits stress-activated protein kinase and cell death, is prepared by the steps of: i) probing proteins binding specifically with stress-activated protein kinase (SAPK) through screening rat's brain cDNA library using yeast two-hybrid system; ii) separating cDNA coding the SAPK specific binding proteins; iii) identifying bindability of SKIP on SAPK; iv) identifying specific bindability of SKIP on SAPK in cultivation cells of mammals; v) identifying bindability of truncated SKIP on SAPK; vi) manufacturing expression vector pcDNA3-rskip-2A and transforming E. coli therewith to obtain E.coli HB101/pcDNA3-rskip-2A(KCTC 8963P); vii) identifying activations of SKIP to control SAPK; and viii) identifying biological characteristics of SKIP able to regulate cell death caused by stress.
