<p>A method for studying the effect of a polymorphism on the activity of a regulatory element such as a promoter is disclosed. The method comprises the steps of providing a first adenoviral vector, comprising: (i) a regulatory element having a first nucleic acid sequence, the regulatory element being operably linked to a reporter gene; (ii) inserting the first adenoviral vector into a suitable host cell; and (iii) detecting a first value of reporter gene expression. This may be compared with the results from a second adenoviral vector which comprises a regulatory element having a different nucleic acid sequence. Alternatively, the first value of reporter gene expression may be compared with that obtained from a vector comprising the promoter-reporter construct attached to a 3'-UTR. This allows the effect of a 3'-UTR on a regulatory element polymorphism to be studied.</p> |
