DEVELOPMENT OF POLYMERASE CHAIN REACTION-SINGLE STRAND CONFORMATION POLYMORPHISM(PCR-SSCP) TO IDENTIFY KOREAN BEEF CATTLE

PURPOSE: Provided is a new technique of polymerase chain reaction-single strand conformation polymorphism(PCR-SSCP) to identify accurately Korean beef cattle in a short time by removing processes which requires expensive restriction enzymes. CONSTITUTION: Polymerase chain reaction-single strand conformation polymorphism(PCR-SSCP) method comprises the following steps of: i) heating at 95 deg.C for 5 minutes MC1R genes multiplied by MC1R gene specific primer 1(forward primer: 5' - CAAGAACCGCAACCTGCACT-3' and reverse primer: 5'-CAAGAACCGCAACCTGCACT-3' and primer 2(forward primer:5'-CAAGAACCGCAACCTGCACT-3' and reverse primer:5'-CCAACGAGGCAGCAGGAT-3'; ii) separating an SSCP band through 12-15 % of unaltered polyacrylamide gel; and iii) dyeing the band with silver.
