The present invention relates to pyruvate decarboxylase (PDC) for transformation of pyruvate in the presence of benzaldehyde to (R)-(-)-phenylacetylcarbinol (product I) of >= 95 % enantiomeric purity with ratio of the product I to 2-hydroxypropiophenone >= 95 % and with specific activity for formation the product I grater than 1U/mg, whose tryptophan radical in position 392 is substituted with a radical of a natural aliphatic amino acid, and further to a process for obtaining pyruvate decarboxylase (PDC) with specific activity greater than 1U/mg for formation of phenylacetylcarbinol, by isolating thereof from a production organism with a gene encoding PDC from Zymomonas mobilis, in the DNA sequence of which the TGG codon in position 1174-1176 encoding for tryptophan radical in position 392 is substituted with a codon encoding a radical of a natural aliphatic amino acid. In the present invention there is also claimed a process of enzymatic preparation of acyloins by enzymatic acyloin condensation of {alpha}-ketocarbonic acids and/or aldehydes in the presence of pyruvate decarboxylase (PDC) whose tryptophan radical in position 392 is substituted with a radical of a natural aliphatic amino acid.
