METHOD FOR ASSAYING THE ACTIVITY OF SIALIDASE

PURPOSE: A method for assaying the activity of sialidase by using enzyme linked immunosorbent assay (ELISA) is provided, thereby the false-positive reaction caused by the residual sialic acid in sample can be inhibited and the activity of sialidase for a target protein can be directly analysed. CONSTITUTION: The method for assaying the activity of sialidase comprises the steps of: fixing sialo glycoprotein(antigen) on the plate attached with sialo glycoprotein antibody; treating with sialidase for an appropriate time and washing sialidase; adding MAA-lectine-digoxigenin(antibody) specifically recognizing sialo oligosaccharide into the sialo glycoprotein and washing to remove the unbinding MAA-lectine-digoxigenin; adding anti-digoxigenin antibody-enzyme conjugate and washing to remove the unbinding conjugate; and adding substrate and measuring the change of color.
