Immobilization of biomolecules on medical devices included converting surface amide groups to amine groups by treatment with hypohalite and hydroxide ions

Producing a medical device comprising biomolecules immobilized on a substrate surface consisting of a polymer having amide functional groups, is new. Producing a medical device comprising biomolecules immobilized on a substrate surface consisting of a polymer having amide functional groups, is new. The method comprises: (a) contacting the polymer with a reaction mixture containing hypohalite and hydroxide ions at at least 20 [deg]C to convert the amide groups to amine groups, where the hydroxide ions are present in a molar excess over the hypohalite ions, and the hydroxide ion concentration in the reaction mixture is no more than 0.1 M; and (b) contacting the amine-functionalized surface with a biomolecule under immobilization conditions. An independent claim is also included for a process for modifying the surface properties of a solid polymeric material, comprising: (a) irradiating the polymeric material surface; (b) contacting the surface with an ethylenically unsaturated amide-functional monomer and oxidizing metal ions to graft the monomer to the surface and form an amide-functional graft copolymer; and (c) carrying out steps (a) and (b) of the novel process.
