PROBLEM TO BE SOLVED: To obtain a new oligonucleotide comprising an oligonucleotide which is targeted to mRNA encoding human A-raf and is capable of inhibiting raf expression, inhibiting hyperproliferation of cells and useful for diagnosis, treatment, etc., of cancer, restenosis, psoriasis, etc. SOLUTION: This new oligonucleotide is targeted to mRNA encoding human A-raf and is capable of inhibiting raf expression and has 8-50 nucleotide length having formula I, II, etc., and modulates expression of the raf gene, a naturally present cellular gene that has been implicated in abnormal cell proliferation and tumor formation and is useful for diagnosis, treatment, etc., of hyperproliferative diseases characterized by activation and growth of cancer, restenosis, psoriasis or T-cell. The oligonucleotide is obtained by designing an oligonucleotide from a base sequence targeting a nucleic acid encoding raf and synthesizing the oligonucleotide on an automated DNA synthesizer by using standard phosphoramidite chemistry. |
