There is provided a plasmid comprising at least one nucleotide sequence that confers to the plasmid, when the plasmid is present in a host cell, the capability of having under certain conditions a controlled, substantially constant low copy number and under different conditions a substantially higher or totally uncontrolled copy number ("runaway" behaviour), and a gene that is expressible in a target cell that is different from the host cell and a method for producing the plasmid. Preferably, the plasmid comprises a sequence coding for a plasmid stabilising gene product. The plasmid is useful in gene therapy including treatment and prevention of infectious diseases such as by vaccination, malignancies and disorders caused by genetically based deficiencies, i.e. genetic diseases. |
