PROBLEM TO BE SOLVED: To obtain a new L-α-glycerophosphate oxidase excellent in reactivity, and useful for clinically determining L-α-glycerophosphate in a serum or the like and for others. SOLUTION: This L-α-glycerophosphate oxidase has the following physicochemical properties. Activity: to form dihydroxy acetone phosphate and hydrogen peroxide by acting on L-α-glycerophosphate in the presence of oxygen; optimal temperature: 35-40 deg.C; optimal pH: 7.5-9.0; thermal stability: not higher than 40 deg.C (treated at pH 7.0 for 15 min); pH stability: stable in the range of 6.0-8.5 (treated at 25 deg.C for 20 hr); molecular weight: 67,000±3000 (SDS-PAGE); and Km value against L-α-glycerophosphate: 3.8±0.5 mM. Further, the L-α- glycerophosphate oxidase is a protein having an amino acid sequence of the formula. The L-α-glycerophosphate oxidase is preferably produced by culturing a transformant prepared by transforming a host cell with a recombinant vector containing a gene encoding the L-α-glycerophosphate oxidase. |
