PROBLEM TO BE SOLVED: To obtain a new plasmid capable of autonomously replicating in Escherichia coli cell and useful for recombinant operation of vector DNA and gene introduction to a bacterium of the genus Gluconobacter by carrying out autonomous replication with a bacterium of the genus Gluconobacter. SOLUTION: This plasmid is an endogenous plasmid derived from Gluconobacter oxydans IFO 3171 strain having about 5.6 Kb size or its derivative and is capable of autonomously replicating with a bacterium of the genus Gluconobacter and has a base sequence expressed by the formula. Furthermore, it is preferable to prepare a shuttle vector built from a plasmid which is either one of pUC18, pSG8 and pSG6 capable of autonomously replicating with the endogenous plasmid or its part and a bacterium of the genus Escherichia, capable of autonomously replicating with a bacterium of the genus Escherichia and capable of autonomously replicating with a bacterium of the genus Gluconobacter and a bacterium of the genus Escherichia, and Escherichia coli JM 109 strain retaining pSG8 is deposited as FERM p-16903. |
