PROBLEM TO BE SOLVED: To obtain a new protein having a specific amino acid sequence, consisting of a protein specifically bindable to an osteoclast formation promoting factor, and useful in searching osteoclast formation inhibitors, as a biochemical reagent, and as a preventive/therapeutic agent for diseases associated with bone metabolism such as osteoporosis. SOLUTION: This new protein is such one as to have an amino acid sequence of the formula and be specifically bindable to an osteoclast formation-promoting factor (OBM), being useful in searching osteoclast formation inhibitors, as a biochemical reagent, and as a preventive/therapeutic agent for diseases associated with bone metabolism such as osteoporosis. This new protein is obtained by the following procedure: the cDNA of OBM is isolated from a cDNA expression library derived from osteoblast-like stroma cells, a gene recombinant-type OBM is prepared using the cDNA, the cDNA library of macrophage-based cell line C7 cells is screened using the gene recombinant-type OBM to obtain a clone capable of producing an OBM-binding protein, and the gene of the clone is integrated into an expression vector and expressed in host cells. |
