PROBLEM TO BE SOLVED: To obtain an isolated DNA molecule useful for the detection of a ketone compound in a biological sample, etc., by adding a structural gene which encodes a protein having the enzymatic activity of 3-hydroxybutyric acid dehydrogenase and which can be obtained from a microorganism. SOLUTION: This DNA molecule contains a structural gene which encodes a protein having an amino acid sequence of the formula and having the enzymatic activity of 3-hydroxybutyric acid dehydrogenase (E.C.1.1.1.30) and which can be obtained from a microorganism such as a bacterium belonging to the genus Rhodobacter, such as Rhodobacter sphaeroides (BMTU 109, DSM 12077). The protein having the enzymatic activity of the 3-hydroxybutyric acid dehydrogenase is preferably prepared by culturing a microorganism, such as Escherichia coli, transformed by the recombined DNA which can be replicated in a microorganism containing both the DNA molecule and a plasmid vector. The protein is further preferably used for the enzymatic measurement of the ketone compound. |
