<p>A process for the preparation of a precipitation antigen Brucella ovis is characterised in that a lyophilised strain Brucella melintensis CAMP 6374 is revived with from 0.3 to 0.5 ml of a liquid cultivating medium (stock). The suspension thus obtained is, after stirring thereof, transferred into 10 ml of a liquid cultivating medium and is further subjected to cultivation together with 5 - 10 percent of CO2, at 37 degrees Celsius, during 5 - 10 days. The liquid cultivating medium is, after incubation thereof, inoculated to an agar medium together with 5 - 10 percent CO2, at 37 degrees Celsius, during 3 - 5 days. After the incubation the typical colonies of the same morphology are skimmed off from the surface of said agar medium, and, bacterial suspension is re-suspended with 100 ml of a liquid suspension medium containing 0.85 percent of sodium chloride or a veronal buffer with pH 7.2. The bacterial suspension thus obtained is stirred and stored in a deep freeze at from -12 to -18 degrees Celsius, for 24 hours after which it is stored in a refrigerator for 24 hours. The whole process of successive freezing and cooling is repeated three times. The bacterial suspension is thereafter treated with ultrasound having the output frequency of 20 kHz during 1 - 3 minutes and is at last stored by means of freezing or conservation.</p> |
