PROBLEM TO BE SOLVED: To obtain MurC polypeptide and MurC polynucleotide useful e.g. for screening of new antibiotics and antibacterial therapies. SOLUTION: This polypeptide has an amino acid sequence at least 70% identical with an amino acid sequence of formula I or II. The other objective polynucleotide includes a polynucleotide sequence encoding a polypeptide having at least 70% identical with the amino acid sequence of formula I or II. The library of a clone, produced e.g. in Escherichia coli, of chromosomal DNA derived from Staphylococcus aureus WCUH 29 is probed by a radio-labeled oligonucleotide derived from a partial sequence of the chromosomal DNA and a clone consisting of the same DNA as the probe is subjected to sequence determination by use of a primer and extended in both the directions so as to determine the whole sequence of the above gene. Polypeptide related to this invention is obtained by culturing host cells subjected to genetic manipulation by a vector including the other objective polynucleotide.
