摘要 PROBLEM TO BE SOLVED: To calibrate/standardize a flow cytometry device with synthetic polymer particles or beads comprising such physical characteristics as provides merit when used with the device. SOLUTION: With calibration or standardization synthetic particles having a specified volume and refraction index, a device which generates light-scatter- related signal comprises (a) process where, with a to-be-assayed particle, calibration or standardization synthetic particle is allowed to pass incident optical flux, (b) process where a first and second measurable signals from calibration or standardization synthetic particles passing the incident optical flux are detected, (c) process where the intensities or the first and second signals are measured to generate a pair of signal intensities, (d) process where the pair of signal intensities is converted into volume and refraction index value and standardized signal gain is given to a flow cytometer, and (e) process where the flow cytometer is calibrated for (1) measurement of volume and refraction index, and (2) counting of analysis-possible particles comprising signal within a range of appropriate volume and refraction index.
