Vector for immuno-toxin expression

Expression vector for producing a recombinant toxin-ligand fusion protein, where the ligand is specific for particular tumour cells, comprises: (a) a sequence coding for a region of an antibody and/or cytokine, (b) a sequence coding for a toxin or the catalytic domain of a toxin, (c) additional sequences for the detection and characterisation of the recombinant protein and (d) unique restriction sites. The vector has a multiple cloning site (MCS) for insertion of antibody variable regions. This is provided with a sequence encoding a (Gly4-Ser)3 linker and is directly compatible with known bacteriophage systems (pCANTAB, pHEN), provided that the unique restriction sites in the MCS are not present in the coding region for the corresponding modified toxin.
