IMPROVEMENT OF TRANSFORMATION OF CULTURED SOYBEAN CELL MEDIATED BY AGROBACTERIUM

PROBLEM TO BE SOLVED: To readily generate a transgenic soybean plant transformed by mediation of Agrobacterium by culturing divided soybean cotyledon node on a nutrient culture medium and culturing the resultant chute in a new nutrient culture medium. SOLUTION: Cotyledon node of soybean is divided and the divided node is cultured until callus tissue containing chute is developed on a nutrient culture medium. Then, the chute is cut off from callus tissue and the chute is rooted on a medium (preferably Gamborg nutrient culture medium) containing hormone and pyroglutamic acid to form a young plant. It is preferable that the young plant is transferred to a culture medium containing soil and grown on the culture medium until a perfect plant is obtained from the young plant. Transformation by Agrobacterium species is preferably carried out by culturing the lower hypocotyl of germinated soybean seed or explant derived from cultured cotyledon node in the presence of a signal compound such as acetosyringone together with Agrobacterium species containing chimeric gene.
