PROBLEM TO BE SOLVED: To obtain an antibody separated and purified from an animal antiserum prepared by using a human recombinant BPGM as an antigen, enabling accurate quantitative determination of bisphosphoglyceromutase(BPGM) and useful e.g. for the diagnosis of hemolytic diseases. SOLUTION: This new antibody against bisphosphoglyceromutase(BPGM) has high specificity, enables accurate quantitative determination of human BPGM participating in the synthesis and decomposition of 2,3-bisphosphoglyceric acid (2,3-BPG) existing in erythrocyte, etc., and is useful e.g. for the diagnosis of arious diseases as a marker, etc., for hemolytic diseases. The antibody against human BPGM can be prepared by producing a human BPGM gene by PCR process using a chromosome DNA originated from human placenta as a template, inserting the gene into an expression vector having tryptophan promoter, expressing the vector in a host cell, administering an animal with the obtained human recombinant BPGM as an antigen, separating the antibody in crude state from the obtained animal antiserum of the immunized animal and purifying the crude antibody. |
