DETECTION OF NUCLEIC ACID USING PROBE

PROBLEM TO BE SOLVED: To provide a detection of nucleic acid using a probe, capable of obtaining an accurate information and detecting nucleic acid without damage to it. SOLUTION: This method for detecting nucleic acid comprises adding a specimen DNA containing a double-stranded DNA 2 bearing a complementary base sequence to a specific base sequence to a mixed solution of a single- stranded DNA probe prepared by cleaving a known specific base sequence between 3'-and 5'-terminals and then combining it with a labeled substance 19 and a Rec A protein molecule 3, moreover superposing a specific base sequence 10 formed at the both terminals of the single-stranded probe on a base sequence of the double-stranded DNA by adding ligase 4, combining the both terminals each other to form a cyclic DNA probe 15 and finally removing Rec A protein molecule to obtain a hybrid DNA. The hybrid DNA is detected by pursuing the labeled substance.
